Separation performance depends on particle size, pore size, column length, and the chemistry of the stationary phase. Smaller particles generally improve efficiency but require higher pressure and suitable instrumentation. The mobile phase often contains buffers and organic solvents that influence retention and selectivity. Testing labs select conditions based on the analytes, sample matrix, and required sensitivity. Method development frequently involves screening several columns and solvent mixtures before a final set of conditions is chosen.
High-performance liquid chromatography is an analytical technique that separates components in a liquid sample by passing them through a packed column under pressure. A pump delivers a mobile phase at a controlled flow rate, and an injector introduces the sample into the stream. Differences in how analytes partition between the mobile phase and the stationary phase cause them to exit the column at different times. Detection then records a signal proportional to the amount of each separated substance. The resulting chromatogram provides retention times and peak areas for identification and quantification.
Method validation examines whether an HPLC procedure is suitable for its intended purpose. Common parameters include accuracy, precision, specificity, linearity, range, detection limit, quantification limit, and robustness. Accuracy describes closeness to a true or accepted value, while precision describes agreement among repeated measurements. Specificity shows whether the method can measure the analyte without interference from related substances. Robustness tests small deliberate changes in flow, temperature, or solvent composition. Validation is not a one-time event; methods may need partial revalidation after changes to instruments, columns, sample handling, or specification limits. Regulatory guidance provides frameworks, but some details remain method-specific.
Regulatory and pharmacopeial texts shape how HPLC testing is performed and documented. The International Council for Harmonisation provides validation guidance, while pharmacopeias publish general chromatography chapters and monographs for specific materials. Accreditation standards such as ISO/IEC 17025 address laboratory competence and traceability. Inspectors may review instrument qualification, analyst training, reference material control, and electronic records. Open questions include how best to validate methods for new complex products and how to handle automated data processing. Laboratories generally resolve these issues through risk assessment, method lifecycle management, and documented scientific justification.
In quality control laboratories, HPLC testing supports batch release, raw material checks, stability studies, and impurity profiling. A validated method defines sample preparation, instrument settings, calibration, and acceptance criteria. Analysts compare results with specifications and investigate out-of-specification outcomes before a batch is approved. Documentation includes chromatograms, integration records, audit trails, and reagent details. Because results influence product decisions, laboratories follow formal quality systems and data integrity rules. The exact tests and limits depend on the material, its intended use, and the applicable regulatory framework.
| Property | Value | Notes |
|---|---|---|
| Separation principle | Differential partitioning | Analytes distribute between mobile and stationary phases. |
| Mobile phase | Liquid solvent mixture | Composition controls retention and selectivity. |
| Stationary phase | Packed column particles | Often chemically bonded silica. |
| Typical detector | UV-Vis or photodiode array | Mass spectrometry is also common. |
| Common synonym | High-performance liquid chromatography | Abbreviated as HPLC. |
Documentation and traceability are central to regulated HPLC testing. Records typically include instrument logs, column history, mobile-phase preparation, sample preparation, injection sequences, raw chromatograms, and audit trails. Electronic systems may require user access controls, time-stamped changes, and backup procedures. Training records show that analysts are qualified for assigned methods. Audits and inspections check whether written procedures match actual practice and whether deviations are documented. These controls support reproducibility and allow results to be reconstructed if questions arise later.
Method validation establishes that an HPLC procedure is suitable for its intended purpose. Typical parameters include accuracy, precision, specificity, linearity, range, limit of detection, limit of quantitation, robustness, and solution stability. Accuracy reflects closeness to a reference value, while precision reflects agreement among repeated measurements. Specificity shows whether the method can measure the analyte without interference from matrix components. Validation is documented through protocols and reports, and the required extent depends on the method's use and regulatory context.
== Translation of NPU definitions == The English NPU definition structure allows for easy and reliable translation of the NPU definitions into other languages; translating each term in the structured NPU definition into another language will produce a valid definition in that language. The concept behind each term in the definition can be determined and agreed on by accessing the international code assigned to that term. The place of the concept in the syntax assigns the specific meaning of the concept in relation to the examination result. Translations of the NPU terminology are directly available in Danish and Swedish. The terminology has been structured such that translations into other languages is straightforward.
Beta blockers, also spelled β-blockers and also sometimes known as β-adrenergic receptor antagonists, are a class of medications predominantly used to manage abnormal heart rhythms (arrhythmia) and to protect the heart from a second heart attack after the first one (secondary prevention). They are also used widely to treat high blood pressure, though they are no longer the first choice for initial treatment of most people. They can also be used to treat anxiety – a notable example being the situational use of propranolol to help dampen the physical symptoms of performance anxiety. Beta blockers are competitive antagonists that block the receptor sites for the endogenous catecholamines epinephrine (adrenaline) and norepinephrine (noradrenaline) on adrenergic beta receptors, of the sympathetic nervous system, which mediates the fight-or-flight response. β-adrenergic receptors are found on cells of the heart muscles, smooth muscles, airways, arteries, kidneys, and other tissues that are part of the sympathetic nervous system and lead to stress responses, especially when they are stimulated by epinephrine (adrenaline). Beta blockers interfere with the binding to the receptor of epinephrine and other stress hormones and thereby weaken the effects of stress hormones. Some beta blockers block activation of all types of β-adrenergic receptors and others are selective for one of the three known types of beta receptors, designated β1, β2, and β3 receptors. β1-Adrenergic receptors are located mainly in the heart and in the kidneys.
In July 2023, when Humira's regulatory exclusivity lapsed, a number of biosimilars such as Hadlima, Hyrimoz, Cyltezo, and Celltrion were launched in the US. In February 2024, adalimumab-ryvk (Simlandi) was approved for medical use in the United States.
On November 28, 2025, The Washington Post published an article alleging Hegseth had given a spoken order to kill the survivors of a September 2, 2025, strike led by SEAL Team 6 on a boat suspected of carrying drugs in the Caribbean Sea. The operation was overseen by Admiral Frank M. "Mitch" Bradley, from Fort Bragg in North Carolina. Bradley described the survivors as legitimate targets, as they could theoretically have contacted other traffickers to rescue them and their cargo, and subsequently ordered the second strike to complete Hegseth's order that everyone must be killed. Hegseth's explanation for the strike has shifted over time. The Office of Legal Counsel eventually argued that the strike was legal because its goal was to destroy the boat, not to kill the crew, reflecting a secret legal memo endorsing such strikes; the memo has been "fiercely criticized" by legal experts. The strike was the first in a series of strikes against suspected drug traffickers. Hegseth's alleged orders were characterized by several legal experts as murder and a war crime. The alleged orders were condemned by members of congress, including Representative Seth Moulton, who said "Mark my words: It may take some time, but Americans will be prosecuted for this, either as a war crime or outright murder." Both Senators Tim Kaine and Mark Kelly described the alleged orders as a war crime.

Slurry ice is a phase changing refrigerant made up of millions of ice "micro-crystals" (typically 0.1 to 1 mm in diameter) formed and suspended within a solution of water and a freezing point depressant. Some compounds used in the field are salt, ethylene glycol, propylene glycol, alcohols like isobutyl and ethanol, and sugars like sucrose and glucose. Slurry ice has greater heat absorption compared to single phase refrigerants like brine, because the melting enthalpy (latent heat) of the ice is also used.
==== Acetone-rosin treatment ==== Acetone-rosin Treatment is sometimes used on dense wood that cannot be penetrated by PEG. This would include softwoods that are nonporous. The goal of this treatment is to replace cells of wood with natural rosin. Rosin is a natural resin that is produced within some woods, for example pines naturally produce resin and are classified as a softwood. As a volatile solvent treatment option, the impact on the health and safety on the workplace must be considered.

== Safety == Cyanuric acid is classified as "essentially nontoxic". The 50% oral median lethal dose (LD50) is 7700 mg/kg in rats. However, when cyanuric acid is present together with melamine (which by itself is another low-toxicity substance), it will form an insoluble and rather nephrotoxic complex, as evidenced in dogs and cats during the 2007 pet food contamination and in children during the 2008 Chinese milk scandal cases.

The treatment for the CSF leak is an extremely invasive procedure that only has a 30% success rate, the family took the chance with the procedure and Lashay had an extremely rough recovery period. Since the treatment none of her symptoms had improved at all. Dr. Lisa Sanders took her story and published it into the Diagnosis column awaiting response from the audience. The audience came up with three general groups of possibilities, she could have a parasitic infection caused by the raccoon attack, POTS, or Rumination syndrome. POTS also known as Postural Orthostatic Tachycardia Syndrome is essentially a problem with the nervous system, specifically the nerves that control the blood vessels causing improper circulation and rapid heart rates resulting in dizziness, light-headedness, and at times vomiting. Rumination syndrome is a rare chronic functional disorder that affects the digestive system where the patient will automatically regurgitate the food and liquids consumed, this syndrome has no cure/treatments. The audience aside from the possible diagnosis also provided Lashay with hope and guidance through tough times. When Dr. Lisa Sanders went to consult with the family about the 3 possibilities, they automatically ruled out both POTS as well as any parasitic infection as they had been tested for almost every kind of parasite as well as received POTS treatment with no effect.
The later demise of the National Assembly proved to be a major setback for Political Catholicism. In the Kingdom of Prussia, the revised constitution of 1850 granted liberties, which in parts even exceeded those of the Frankfurt draft constitution, yet two years later the minister for culture, von Raumer, issued decrees directed mainly against the Jesuits. In reaction this led to a doubling of Catholic representatives in the subsequent elections and the formation of a Catholic club in the Landtag of Prussia. In 1858, when the "New Era" governments of Wilhelm I adopted more lenient policies, the club renamed itself "Fraction of the Centre" in order to open itself up to include non-Catholics. This name stemmed from the fact that in the Landtag the Catholic representatives were seated in the centre, between the Conservatives on the right and the Liberals on the left. Faced with military and constitutional issues, where there was no definite Church position, the group soon disintegrated and disappeared from parliament after 1867.
==== Quantum dot in hybrid solar cells ==== Colloidal quantum dots are also used in inorganic–organic hybrid solar cells. These solar cells are attractive because of the potential for low-cost fabrication and relatively high efficiency. Incorporation of metal oxides, such as ZnO, TiO2, and Nb2O5 nanomaterials into organic photovoltaics have been commercialized using full roll-to-roll processing. A 13.2% power conversion efficiency is claimed in Si nanowire/PEDOT:PSS hybrid solar cells.
== Arab counter-proposal == On 21 February, the Arab leaders of Egypt, Jordan, Qatar, UAE, Saudi Arabia, Kuwait, and Bahrain met in Saudi Arabia to discuss the future of Gaza. Their goal was to be a "united front" and provide an alternative to Trump's plan. Ibrahim Dalalsha, director of a political research group in West Bank, said the process would be complex and that the greatest obstacle for the Arab leaders is to introduce a plan that is realistic and can be enforced on the Palestinian factions, while at the same time being a plan that the US and Israel could accept. Arab leaders planned to meet for a broader summit on 4 March in Cairo to find a plan for Gaza before their diplomats travel to Washington D.C. "within weeks" to meet with President Trump to present their counter plan.

HPLC separates and detects individual compounds in a liquid sample, producing peaks at characteristic retention times. Peak area or height can be used to estimate concentration when calibrated with known standards. It does not identify unknown compounds with certainty unless additional detectors or reference materials are used.
Pressure drives the liquid mobile phase through a column packed with small particles. Without pressure, flow would be very slow or stop because the packed bed resists liquid movement. Modern pumps maintain a steady flow despite the resistance.
A chromatogram is a plot of detector signal against time after sample injection. Each peak represents a compound or group of compounds eluting from the column. Retention time and peak area are the main measurements read from the plot.
System suitability is a set of checks performed before and during an HPLC run to confirm that the instrument and method are working as expected. It may include retention time repeatability, resolution between peaks, peak symmetry, and signal intensity. Failing suitability criteria usually invalidates the run.
