Developing an HPLC method begins with defining the purpose, such as quantifying a main component, measuring impurities, or confirming identity. Analysts select separation mode, column, mobile phase, detection, and sample preparation based on analyte properties and matrix. Experiments vary solvent strength, pH, buffer type, and temperature to achieve resolution between critical peaks. The goal is a robust method that produces reliable results across instruments and operators. Method development often involves trial runs and statistical optimization.
Validation demonstrates that a method is suitable for its intended use. Typical performance characteristics include accuracy, precision, specificity, linearity, range, limit of detection, limit of quantitation, and robustness. Regulators and standards organizations provide frameworks, but specific requirements depend on the application and jurisdiction. System suitability tests are run before sample analysis to confirm resolution, peak symmetry, retention time repeatability, and sensitivity. A validated method is not permanently fixed; changes may require partial or full revalidation.
Routine HPLC testing depends on controlled reagents, calibrated instruments, and documented procedures. Columns degrade over time, so retention times and peak shapes are monitored for drift. Mobile phases are filtered and degassed to prevent pump damage and detector noise. Reference standards must be traceable and stored under suitable conditions. Data handling systems record injections, calculations, and audit trails. Quality control samples interspersed with unknowns help detect errors during a run.
System suitability testing is performed before and during analytical runs to confirm that the instrument and method are working as expected. Typical checks include retention time, peak area precision, resolution between critical pairs, tailing factor, and theoretical plate count. Acceptance criteria are set in the method or pharmacopeial monograph. If a suitability check fails, the run may be rejected and the instrument or sample preparation may need investigation. This practice helps prevent release of data from a system that has drifted out of control.
Quality control samples are inserted at intervals to monitor accuracy and precision throughout a batch. Blank samples detect contamination, while spiked samples assess recovery from the sample matrix. Calibration standards establish the relationship between detector response and concentration, and control samples are prepared independently from them whenever possible. Laboratories also participate in proficiency testing and maintain audit trails, instrument logs, and reagent records. Ongoing review of control charts can reveal trends before they cause out-of-specification results.
Method validation demonstrates that an HPLC procedure is suitable for its intended purpose. Common validation parameters include accuracy, precision, specificity, linearity, range, limit of detection, limit of quantification, and robustness. Accuracy reflects agreement with a reference value, while precision describes repeatability under defined conditions. Specificity shows whether the method can measure the analyte in the presence of impurities or matrix components. Validation documents are reviewed before a method is used for routine testing or regulatory submissions.
| Property | Value | Notes |
|---|---|---|
| Validation parameter | Accuracy | Closeness of measured value to accepted reference value |

| System suitability check | Resolution ≥ 1.5 | Baseline separation between critical peak pair |
| System suitability check | Tailing factor ≤ 2.0 | Common target for peak symmetry |
| Documentation | Validation report | Summarizes experiments, acceptance criteria, and conclusions |
Documentation and traceability are central to regulated HPLC testing. Records typically include instrument logs, column history, mobile-phase preparation, sample preparation, injection sequences, raw chromatograms, and audit trails. Electronic systems may require user access controls, time-stamped changes, and backup procedures. Training records show that analysts are qualified for assigned methods. Audits and inspections check whether written procedures match actual practice and whether deviations are documented. These controls support reproducibility and allow results to be reconstructed if questions arise later.
Method validation establishes that an HPLC procedure is suitable for its intended purpose. Typical parameters include accuracy, precision, specificity, linearity, range, limit of detection, limit of quantitation, robustness, and solution stability. Accuracy reflects closeness to a reference value, while precision reflects agreement among repeated measurements. Specificity shows whether the method can measure the analyte without interference from matrix components. Validation is documented through protocols and reports, and the required extent depends on the method's use and regulatory context.
Routine quality control uses system suitability, blank injections, check standards, and control samples to detect drift or contamination. System suitability criteria may specify minimum resolution, maximum tailing factor, and a permitted range for repeated injections. Blank injections reveal carryover or solvent contamination, while check standards confirm calibration accuracy over a batch. Control samples with known analyte levels can show whether results remain within statistical limits. When a control result falls outside limits, the analyst investigates the cause and may invalidate affected results before repeating the batch.
Routine HPLC testing compares a sample result with a calibration curve prepared from known reference standards. Peak area or peak height is plotted against concentration, and the curve is used to estimate unknown amounts. Retention time supports tentative identification when compared with a standard, though mass spectrometry or another confirmatory method may be needed for definitive identification. Pre-run checks verify repeatability, resolution, and peak symmetry before sample analysis. Limits of detection and quantification describe the smallest amounts that can be reliably observed or measured. Sample preparation, filtration, and degassing help prevent column damage and inconsistent results.
High-performance liquid chromatography is an analytical technique that separates components in a liquid sample. A pump moves a liquid mobile phase through a column packed with a solid stationary phase. Compounds interact differently with both phases and travel at different rates, leaving the column at distinct retention times. A detector records these arrivals as peaks on a chromatogram. The resulting pattern supports identification and quantification of substances in mixtures. Modern instruments use high pressure to force solvent through small particles, which improves speed and resolution compared with older low-pressure liquid chromatography methods.
After the plebiscite in Masuria, attacks on Poles occurred by pro-German mobs. In particular, Polish priests and politicians were charged, even in their homes. These results were published by Poland in 1920, but gave Polish place names as fixed in the late 1940s:
reassociation kinetics The measurement and manipulation of the rate of reannealing of complementary strands of DNA, generally by heating and denaturing a double-stranded molecule into single strands and then observing their rehybridization at a cooler temperature. Because the base pair G+C requires more energy to anneal than the base pair A+T, the rate of reannealing between two strands depends partly on their nucleotide sequence, and it is therefore possible to predict or estimate the sequence of the duplex molecule by the time it takes to fully hybridize. Reassociation kinetics is studied with C0t analysis: fragments reannealing at low C0t values tend to have highly repetitive sequences, while higher C0t values imply more unique sequences.
Romanian cuisine has been influenced by Austrian and German cuisine (especially in the historical regions that had been formerly administered by the Habsburg monarchy), but also shares some similarities with other cuisines in the Balkan region such as Greek, Bulgarian, and Serbian cuisine. Ciorbă includes a wide range of sour soups, while mititei, mămăligă (similar to polenta), and sarmale are featured commonly in main courses. Pork, chicken, and beef are the preferred types of meat, but lamb and fish are also quite popular. Certain traditional recipes are made in direct connection with the holidays: chiftele, tobă and tochitură at Christmas; drob, pască and cozonac at Easter and other Romanian holidays. Țuică is a strong plum brandy reaching a 70% alcohol content which is the country's traditional alcoholic beverage, taking as much as 75% of the national crop (Romania is one of the largest plum producers in the world). Traditional alcoholic beverages also include wine, rachiu, palincă and vișinată, but beer consumption has increased dramatically over recent years. In 2019, Romania became the country with the highest alcohol consumption per capita.
Charnia fossils were originally found in the Charnwood Forest in England, hence named Charnia. These fossils are from marine organisms that lived on the bottom of the ocean floor. The fossils have a fractal body plan and were frond shaped, meaning they resembled broad-leafed plants such as ferns. However they could not have been plants since they resided in the dark depths of the ocean floor. In Charnwood Forest, Charnia was found as an isolated species, however there were many more fossils found on the East Coast of Canada in Mistaken Point in Newfoundland. Charnia was attached to the bottom of the ocean floor, and was strongly current aligned. This is seen because there are disk-like shapes at the bottom of the Charnia fossil, which show where Charnia was tethered, and all the nearby fossils are facing the same direction. These fossils at Mistaken Point were preserved well under volcanic ash and layers of soft mud. It has been determined via radiometric dating of the fossils that Charnia must have lived around 565 million years ago.

== Epidemiology == The exact number of cases of rhabdomyolysis is difficult to establish because different definitions have been used. In 1995, hospitals in the U.S. reported 26,000 cases of rhabdomyolysis. Up to 85% of people with major traumatic injuries will experience some degree of rhabdomyolysis. Of those with rhabdomyolysis, 10–50% develop acute kidney injury. The risk is higher in people with a history of illicit drug use, alcohol misuse, or trauma when compared to muscle diseases, and it is particularly high if multiple contributing factors occur together. Rhabdomyolysis accounts for 7–10% of all cases of acute kidney injury in the U.S. Crush injuries are common in major disasters, especially in earthquakes. The aftermath of the 1988 Spitak earthquake prompted the establishment, in 1995, of the Renal Disaster Relief Task Force, a working group of the International Society of Nephrology (a worldwide body of kidney experts). Its volunteer doctors and nurses assisted for the first time in the 1999 İzmit earthquake in Turkey, where 17,480 people died, 5392 were hospitalized, and 477 received dialysis, with positive results. Treatment units are generally established outside the immediate disaster area, as aftershocks could potentially injure or kill staff and make equipment unusable. Acute exertional rhabdomyolysis happens in 2% to 40% of people going through basic training for the United States military. In 2012, the United States military reported 402 cases. Another group at increased risk is firefighters.
== Early years == Mariusz Pudzianowski was born in Biała Rawska, Poland. His father, Wojciech, was a weightlifter. Pudzianowski quickly became interested in sports. Since the age of 11, he has been training the Kyokushin style of karate. His current grade is 4th kyu green belt. He began strength training at the age of thirteen. When he was fifteen, Pudzianowski also started training boxing, quitting after seven years. Pudzianowski debuted in professional sports at the age of sixteen, taking part in Polish Weightlifting Championship, in the bench press event.
== History == Evidence for periostitis found in the fossil record is studied by paleopathologists, specialists in ancient disease and injury. Periostitis has been seen in the late Cretaceous-Eocene crocodile Borealosuchus formidabilis, once known as Leidyosuchus. In one study, periostitis was the most common pathology in this species, with 134 instances of the condition out of 7,154 bones the scientists examined showing evidence for the condition. Periostitis has also been documented in dinosaurs, including a forelimb referred to as the long-necked Camarasaurus grandis, as well as the shoulder blade of a horned dinosaur.
Justin McNulty is suspended from the SDLP for leaving the Assembly sitting early to manage Laois GAA at a Gaelic football match in Wexford. Sir Bob Neill, MP for Bromley and Chislehurst, announces he is stand down at the next election to spend more time with his wife, who has suffered a stroke. 4 February – Dafydd Wigley, a former leader of Plaid Cymru, warns that reforms to Wales's political system pose "a very great danger" since it would destroy the relationship between voters and the people they elect. Prime Minister Rishi Sunak arrives in Northern Ireland to visit ministers following the restoration of the Executive. 5 February – Sunak visits Stormont along with Taoiseach Leo Varadkar to mark the restoration of the Northern Ireland Executive. Sunak is criticised by opposition parties after appearing to agree to a £1,000 bet on the Rwanda asylum plan, that the first flight to Rwanda would take off before the election. He subsequently says the challenge, put forward by TalkTV presenter Piers Morgan, took him by surprise, but that it was not a mistake to accept it. Sunak says the government has "not made enough progress" on cutting NHS waiting lists in England, but that industrial action "has had an impact". The UK government sets out its Disability Action Plan, which includes measures to protect people with assistance dogs from being illegally refused entry to businesses. The UK government launches a six-week consultation on plans for Martyn's Law, which would make provisions to better protect the public against potential acts of terrorism.
=== Production during the Manhattan Project === During World War II the U.S. government established the Manhattan Project, for developing an atomic bomb. The three primary research and production sites of the project were the plutonium production facility at what is now the Hanford Site, the uranium enrichment facilities at Oak Ridge, Tennessee, and the weapons research and design lab, now known as Los Alamos National Laboratory, LANL.

=== Abiotic stress resistance === Overexpression of systemin and HypSys has been found to improve plants' tolerance to abiotic stress, including salt stress and UV radiation. When prosystemin was over-expressed in tomato, transgenic plants had lower stomatal conductance than normal plants. When grown in salt solutions, transgenic plants had higher stomatal conductances, lower leaf concentrations of abscisic acid and proline and a higher biomass. These findings suggest that systemin either allowed the plants to adapt to salt stress more efficiently or that they perceived a less stressful environment. Similarly, wounded tomato plants were less susceptible to salt stress than unwounded plants. This may be because wounding decreases the growth of the plant and therefore slows the uptake of toxic ions into the roots. An analysis of salt-induced changes in gene expression found that the differences measured between the transgenic and normal plants could not be accounted for by changes in conventional salt stress-induced pathways. Instead Orsini et al. suggested that the activation of the jasmonic acid pathway determines a physiological state that not only directs resources towards the production of compounds active against pests, but also pre-adapts plants to minimize water loss. These effects are achieved by negatively regulating the production of hormones and metabolites that will force plants to invest additional resources to counteract water loss, a secondary effect of herbivores.
In her reply, Meitner concurred. "At the moment, the interpretation of such a thoroughgoing breakup seems very difficult to me, but in nuclear physics we have experienced so many surprises, that one cannot unconditionally say: 'it is impossible'." On 22 December 1938, Hahn sent a manuscript to Naturwissenschaften reporting their radiochemical results, which were published on 6 January 1939. On 27 December, Hahn telephoned the editor of the Naturwissenschaften and requested an addition to the article, speculating that some platinum group elements previously observed in irradiated uranium, which were originally interpreted as transuranium elements, could in fact be technetium (then called "masurium"), mistakenly believing that the atomic masses had to add up rather than the atomic numbers. By January 1939, he was sufficiently convinced of the formation of light elements that he published a new revision of the article, retracting former claims of observing transuranic elements and neighbours of uranium. As a chemist, Hahn was reluctant to propose a revolutionary discovery in physics, but Meitner and Frisch worked out a theoretical interpretation of nuclear fission, a term appropriated by Frisch from biology. In January and February they published two articles discussing and experimentally confirming their theory.
=== New Zealand === Beak trimming, officially known as 'beak tipping' in law, is legal but regulated in New Zealand. The Layer Hens Code of Welfare (last updated 1 October 2018) notes that the National Animal Welfare Advisory Committee (NAWAC) "encourages the industry to develop management systems to protect against all forms of injurious pecking without the need for beak tipping". The Code recommends that "[a]lternative strategies for managing injurious (feather) pecking that minimise the need for beak tipping should be employed e.g. use and availability of different foraging resources." In case that beak tipping is deemed necessary, however, it requires competent trained operators, must be carried out within 3 days of hatching (except in emergencies under veterinary supervision) and may not remove more than one-quarter of the upper or lower beaks.
Vasoconstriction (narrowing of blood vessels) and vascular smooth muscle hypertrophy (enlargement) induced by ATII may lead to increased blood pressure and hypertension. Further, constriction of the efferent arterioles of the kidney leads to increased perfusion pressure in the glomeruli. It contributes to ventricular remodeling and ventricular hypertrophy of the heart through stimulation of the proto-oncogenes c-fos, c-jun, c-myc, transforming growth factor beta (TGF-B), through fibrogenesis and apoptosis (programmed cell death). Stimulation by ATII of the adrenal cortex to release aldosterone, a hormone that acts on kidney tubules, causes sodium and chloride ions retention and potassium excretion. Sodium is a "water-holding" ion, so water is also retained, which leads to increased blood volume, hence an increase in blood pressure. Stimulation of the posterior pituitary to release vasopressin (antidiuretic hormone, ADH) also acts on the kidneys to increase water retention. If ADH production is excessive in heart failure, Na+ level in the plasma may fall (hyponatremia), and this is a sign of increased risk of death in heart failure patients. A decrease renal protein kinase C During the course of ACE inhibitor use, the production of ATII is decreased, which prevents aldosterone release from the adrenal cortex. This allows the kidney to excrete sodium ions along with water, and retain potassium ions. This decreases blood volume, leading to decreased blood pressure.
Spironolactone is not generally used in men for the treatment of androgen-dependent dermatological conditions because of its feminizing side effects, but it is effective for such indications in men similarly. As an example, spironolactone has been reported to reduce symptoms of acne in males. An additional example is the usefulness of spironolactone as an antiandrogen in transgender women and nonbinary individuals. Topical spironolactone is effective in the treatment of acne, as well. As a result, topical pharmaceutical formulations containing 2 or 5% spironolactone cream became available in Italy for the treatment of acne and hirsutism in the early 1990s. The products were discontinued in 2006 when the creams were added to the list of doping substances with a decree of the Ministry of Health that year.

It is a set of checks performed before or during an HPLC run to confirm the system works as expected. Parameters may include resolution, tailing factor, theoretical plates, and retention time precision. Failure can trigger maintenance, method adjustment, or repeat analysis.
Validation follows a planned protocol that tests accuracy, precision, specificity, linearity, range, detection limits, quantitation limits, and robustness. Results are compared against predefined acceptance criteria. The validation report supports regulatory filing or routine use.
Revalidation may be needed after changes to column chemistry, mobile phase, detection, sample preparation, or instrument type. It can also follow a pattern of out-of-specification results. The scope depends on whether the change affects method performance.
Method validation is the documented process of showing that an HPLC procedure produces reliable results for a defined purpose. It examines parameters such as accuracy, precision, specificity, linearity, and robustness. Regulators and quality systems often require validation before routine use.
