Practical HPLC testing depends on careful sample preparation and instrument maintenance. Samples may require filtration, dilution, pH adjustment, or extraction to avoid column damage and matrix interference. Mobile phases are degassed and filtered, and columns are equilibrated before injection. Common problems include peak tailing, baseline drift, ghost peaks, carryover, and co-elution of analytes. Documentation of instrument logs, calibration records, and electronic audit trails supports data integrity and traceability. Ongoing training and routine maintenance help reduce variability between analysts and laboratories.
Quality control laboratories use HPLC to check identity, purity, concentration, and stability of raw materials and finished products. A validated method specifies the column, mobile phase, flow rate, detection wavelength, injection volume, and run time. Samples are prepared and compared against reference standards of known concentration. The resulting chromatogram provides quantitative data, such as assay values and impurity levels. This approach is common in pharmaceutical, food, environmental, and industrial testing where consistent measurements are required.
Instrumentation includes a solvent delivery system, an autosampler, a column oven, and one or more detectors. Reversed-phase columns with chemically modified silica are widely used, but normal-phase, ion-exchange, size-exclusion, and affinity modes exist for specific separations. Detectors may rely on ultraviolet absorbance, fluorescence, refractive index, or mass spectrometry. Column temperature, mobile phase composition, and flow rate are adjusted to improve resolution. System pressure is monitored because rising pressure can indicate column blockage or deteriorating packing.
Separation performance depends on particle size, pore size, column length, and the chemistry of the stationary phase. Smaller particles generally improve efficiency but require higher pressure and suitable instrumentation. The mobile phase often contains buffers and organic solvents that influence retention and selectivity. Testing labs select conditions based on the analytes, sample matrix, and required sensitivity. Method development frequently involves screening several columns and solvent mixtures before a final set of conditions is chosen.
| Parameter | Typical acceptance criterion | Notes |
|---|---|---|
| Resolution | ≥ 1.5 | Baseline separation of adjacent peaks |
| Tailing factor | ≤ 2.0 | Peak symmetry measure |
| Theoretical plates | > 2000 | Column efficiency indicator |
| Injection repeatability | ≤ 2% RSD | Relative standard deviation for replicate injections |
| Linearity | r² ≥ 0.995 | Calibration curve over the working range |
Routine HPLC testing compares a sample result with a calibration curve prepared from known reference standards. Peak area or peak height is plotted against concentration, and the curve is used to estimate unknown amounts. Retention time supports tentative identification when compared with a standard, though mass spectrometry or another confirmatory method may be needed for definitive identification. Pre-run checks verify repeatability, resolution, and peak symmetry before sample analysis. Limits of detection and quantification describe the smallest amounts that can be reliably observed or measured. Sample preparation, filtration, and degassing help prevent column damage and inconsistent results.
High-performance liquid chromatography is an analytical technique that separates components in a liquid sample. A pump moves a liquid mobile phase through a column packed with a solid stationary phase. Compounds interact differently with both phases and travel at different rates, leaving the column at distinct retention times. A detector records these arrivals as peaks on a chromatogram. The resulting pattern supports identification and quantification of substances in mixtures. Modern instruments use high pressure to force solvent through small particles, which improves speed and resolution compared with older low-pressure liquid chromatography methods.
Routine quality control monitors retention time shifts, baseline noise, system pressure, and peak shape. Trends can reveal column aging, mobile phase preparation errors, detector drift, or sample degradation. Corrective actions may include replacing the column, preparing fresh mobile phase, or recalibrating the detector. Stability testing often uses HPLC to measure parent compound loss and degradation product formation. Open questions remain about how accelerated stability results extrapolate to long-term storage under varied conditions.
Quality control for HPLC testing combines scheduled checks, documented procedures, and review of results. Before sample analysis, system suitability testing confirms that the instrument, column, and method meet predefined criteria. Common criteria include resolution between critical peaks, retention time precision, peak tailing, and theoretical plate count. Failure triggers investigation before results are reported. Records link raw data, calculations, instrument logs, and analyst identity to each batch, supporting audits and repeat analysis.
Validation demonstrates that a method is suitable for its intended use. Typical performance characteristics include accuracy, precision, specificity, linearity, range, limit of detection, limit of quantitation, and robustness. Regulators and standards organizations provide frameworks, but specific requirements depend on the application and jurisdiction. System suitability tests are run before sample analysis to confirm resolution, peak symmetry, retention time repeatability, and sensitivity. A validated method is not permanently fixed; changes may require partial or full revalidation.
Routine HPLC testing depends on controlled reagents, calibrated instruments, and documented procedures. Columns degrade over time, so retention times and peak shapes are monitored for drift. Mobile phases are filtered and degassed to prevent pump damage and detector noise. Reference standards must be traceable and stored under suitable conditions. Data handling systems record injections, calculations, and audit trails. Quality control samples interspersed with unknowns help detect errors during a run.
Developing an HPLC method begins with defining the purpose, such as quantifying a main component, measuring impurities, or confirming identity. Analysts select separation mode, column, mobile phase, detection, and sample preparation based on analyte properties and matrix. Experiments vary solvent strength, pH, buffer type, and temperature to achieve resolution between critical peaks. The goal is a robust method that produces reliable results across instruments and operators. Method development often involves trial runs and statistical optimization.
=== In sport === The WADA has prohibited the use of tramadol in competition since 1 January 2024. Misuse of painkillers is up to four times more common among athletes compared to the general population. From March 2019, the Union Cycliste Internationale (UCI) banned the drug, after riders were using the painkiller to improve their performance. No significant improvements in performance were found when compared to placebo. In 2026 the International Go Federation (IGF) revoked Korean player Sa Woo Kim’s third place in the 2025 World Amateur Go Championship in Vancouver, Canada because he had tested positive for tramadol. The IGF adopted the World Anti-Doping Code in October 2020.

GLP-1 possesses several physiological properties making it (and its functional analogs) a subject of intensive investigation as a potential treatment of diabetes mellitus, as these actions induce long-term improvements along with the immediate effects. Although reduced GLP-1 secretion has previously been associated with attenuated incretin effect in patients with type 2 diabetes, further research indicates that GLP-1 secretion in patients with type 2 diabetes does not differ from healthy subjects. The most noteworthy effect of GLP-1 is its ability to promote insulin secretion in a glucose-dependent manner. As GLP-1 binds to GLP-1 receptors expressed on pancreatic β cells, the receptors couple to G-protein subunits and activate adenylate cyclase, which increases the production of cAMP from ATP. Subsequently, activation of secondary pathways, including protein kinase A (PKA) and Epac2, alters cell ion channel activity, causing elevated levels of cytosolic Ca2+ that enhance exocytosis of insulin-containing granules. During the process, influx of glucose ensures sufficient ATP to sustain the stimulatory effect. Additionally, GLP-1 ensures the β cell insulin stores are replenished to prevent exhaustion during secretion by promoting insulin gene transcription, mRNA stability and biosynthesis. GLP-1 also increases β cell mass by promoting proliferation and neogenesis while inhibiting apoptosis. As both type 1 and 2 diabetes are associated with reduction of functional β cells, this effect is desirable in diabetes treatment.

=== Differential diagnosis === An important differential diagnosis of OI is child abuse, as both may present to a clinician with multiple fractures in various stages of healing. Differentiating them can be difficult, especially when no other characteristic features of OI are present. This can become an issue in court; in the United States, several child abuse cases were resolved with a finding that osteogenesis imperfecta was the true cause of a child's fractures, leading to lawsuits seeking redress such as Alice Velasquez, et al. v. United States. Other differential diagnoses include rickets and osteomalacia, both caused by malnutrition, as well as rare skeletal/connective tissue syndromes such as Bruck syndrome, hypophosphatasia, geroderma osteodysplasticum, and Ehlers–Danlos syndrome. Various forms of osteoporosis, such as iatrogenic osteoporosis, idiopathic juvenile osteoporosis, disuse osteoporosis and exercise-related osteoporosis should also be considered when OI is suspected.
== Applications == The main application of 238Pu is as the heat source in radioisotope thermoelectric generators (RTGs). The RTG was invented in 1954 by Mound scientists Ken Jordan and John Birden, who were inducted into the National Inventors Hall of Fame in 2013. They immediately produced a working prototype using a 210Po heat source, and on January 1, 1957, entered into an Army Signal Corps contract (R-65-8- 998 11-SC-03-91) to conduct research on radioactive materials and thermocouples suitable for the direct conversion of heat to electrical energy using polonium-210 as the heat source. In 1966, a study reported by SAE International described the potential for the use of plutonium-238 in radioisotope power subsystems for applications in space. This study focused on employing power conversions through the Rankine cycle, Brayton cycle, thermoelectric conversion, and thermionic conversion with plutonium-238 as the primary heating element. The heat supplied by the plutonium-238 heating element was consistent between the 400 °C and 1000 °C regime but future technology could reach an upper limit of 2000 °C, further increasing the efficiency of the power systems. The Rankine cycle study reported an efficiency between 15 and 19% with inlet turbine temperatures of 730 °C, whereas the Brayton cycle offered efficiency greater than 20% with an inlet temperature of 840 °C. Thermoelectric converters offered low efficiency (3-5%) but high reliability. Thermionic conversion could provide similar efficiencies to the Brayton cycle if proper conditions reached.
The Trump administration took the unusual step of subjecting approximately 50 members of the US military's Joint Chiefs of Staff to polygraph tests over leaks to journalists, including information about how the war has resulted in a shortage of critical munitions. Early attacks on US bases caused logistics issues in US aircraft carrier USS Abraham Lincoln. The Pentagon has transferred weapons stored in Asia and Europe to replenish stockpiles in the Middle East. Still, missile interceptors shortage reportedly persuaded Trump to call off a planned major attack on Iran. The Trump administration blames the shortage on Joe Biden giving too many weapons to Ukraine. On 17 September, Lockheed Martin received the first batch of Patriot interceptor parts from General Motors less than a month after the two companies signed a manufacturing agreement, in a success for the Pentagon's push for faster weapon production, particularly the Patriot missile defense system, as it remained one of the world's most sought-after weapons for the war and Russia's war in Ukraine.
A study on Neanderthal teeth from the Chagyrskaya Cave in the Altai Mountains (Russia), indicating that the studied sample overall falls within the known Neanderthal phenotypic variability but also preserves specific morphological traits, is published by Gicqueau et al. (2026). Massilani et al. (2026) present a high-quality genome of an approximately 110,000-years-old Neanderthal individual from the Denisova Cave (Russia), providing evidence of a closer relationship of the studied individual to a 120,000-years-old Neanderthal from the same cave than to a 80,000-years-old individual from the Chagyrskaya Cave or to European Neanderthals, evidence of gene flow from Denisovans in both Neanderthals from the Denisova Cave, and evidence of differentiation between Altai and European Neanderthals comparable to that of the most differentiated populations of modern humans. Zubova et al. (2026) report evidence of deliberate invasive dental caries intervention in a 59,000-years-old Neanderthal molar from the Chagyrskaya Cave. Evidence from the study of rhinocerotid remains from Middle Paleolithic sites in France and Spain, interpreted as consistent with use of rhinoceros teeth as tools (including soft hammers and anvils) by Neanderthals, is presented by Sanz-Royo et al. (2026). Picin et al.
=== Layers === The peritoneum is one continuous sheet, forming two layers and a potential space between them: the peritoneal cavity. The outer layer, the parietal peritoneum, is attached to the abdominal wall and the pelvic walls. The tunica vaginalis, the serous membrane covering the male testis, is derived from the vaginal process, an outpouching of the parietal peritoneum. The inner layer, the visceral peritoneum, is wrapped around the visceral organs, located inside the intraperitoneal space for protection. It is thinner than the parietal peritoneum. The mesentery is a double layer of visceral peritoneum that attaches to the gastrointestinal tract. There are often blood vessels, nerves, and other structures between these layers. The space between these two layers is technically outside of the peritoneal sac, and thus not in the peritoneal cavity. The potential space between the parietal and visceral peritoneum is the peritoneal cavity, filled with a small amount (about 50 mL) of slippery serous fluid that allows the two layers to slide freely over each other. The right paracolic gutter is continuous with the right and left subhepatic spaces. The epiploic foramen allows communication between the greater sac and the lesser sac. The peritoneal space in males is closed, while the peritoneal space in females is continuous with the extraperitoneal pelvis through openings of the fallopian tubes, the uterus, and the vagina.

Method validation is the documented process of confirming that an HPLC procedure is suitable for its intended use. It evaluates accuracy, precision, specificity, linearity, range, detection limits, and robustness. Validation criteria depend on the regulatory context and the sample type.
System suitability tests are short checks performed before or during an HPLC run to verify instrument and method performance. They often include resolution, tailing factor, theoretical plates, and injection precision. Results must meet predefined limits for sample data to be accepted.
HPLC retention time alone cannot definitively identify an unknown substance. A match with a reference standard under identical conditions provides supporting evidence. Confirmation typically requires mass spectrometry, nuclear magnetic resonance, or another orthogonal technique.
HPLC separates and detects individual compounds in a liquid sample, producing peaks at characteristic retention times. Peak area or height can be used to estimate concentration when calibrated with known standards. It does not identify unknown compounds with certainty unless additional detectors or reference materials are used.
