High-performance liquid chromatography is an analytical technique that separates components in a liquid sample by passing them through a packed column under pressure. A pump delivers a mobile phase at a controlled flow rate, and an injector introduces the sample into the stream. Differences in how analytes partition between the mobile phase and the stationary phase cause them to exit the column at different times. Detection then records a signal proportional to the amount of each separated substance. The resulting chromatogram provides retention times and peak areas for identification and quantification.
Instrumentation includes a solvent delivery system, an autosampler, a column oven, and one or more detectors. Reversed-phase columns with chemically modified silica are widely used, but normal-phase, ion-exchange, size-exclusion, and affinity modes exist for specific separations. Detectors may rely on ultraviolet absorbance, fluorescence, refractive index, or mass spectrometry. Column temperature, mobile phase composition, and flow rate are adjusted to improve resolution. System pressure is monitored because rising pressure can indicate column blockage or deteriorating packing.
Routine quality control uses system suitability, blank injections, check standards, and control samples to detect drift or contamination. System suitability criteria may specify minimum resolution, maximum tailing factor, and a permitted range for repeated injections. Blank injections reveal carryover or solvent contamination, while check standards confirm calibration accuracy over a batch. Control samples with known analyte levels can show whether results remain within statistical limits. When a control result falls outside limits, the analyst investigates the cause and may invalidate affected results before repeating the batch.
Documentation and traceability are central to regulated HPLC testing. Records typically include instrument logs, column history, mobile-phase preparation, sample preparation, injection sequences, raw chromatograms, and audit trails. Electronic systems may require user access controls, time-stamped changes, and backup procedures. Training records show that analysts are qualified for assigned methods. Audits and inspections check whether written procedures match actual practice and whether deviations are documented. These controls support reproducibility and allow results to be reconstructed if questions arise later.
Method validation establishes that an HPLC procedure is suitable for its intended purpose. Typical parameters include accuracy, precision, specificity, linearity, range, limit of detection, limit of quantitation, robustness, and solution stability. Accuracy reflects closeness to a reference value, while precision reflects agreement among repeated measurements. Specificity shows whether the method can measure the analyte without interference from matrix components. Validation is documented through protocols and reports, and the required extent depends on the method's use and regulatory context.
| Property | Value | Notes |
|---|---|---|

| Mobile phase | Liquid solvent mixture | Composition controls retention and selectivity. |
| Stationary phase | Packed column particles | Often chemically bonded silica. |
| Typical detector | UV-Vis or photodiode array | Mass spectrometry is also common. |
| Common synonym | High-performance liquid chromatography | Abbreviated as HPLC. |
Regulatory and pharmacopeial texts shape how HPLC testing is performed and documented. The International Council for Harmonisation provides validation guidance, while pharmacopeias publish general chromatography chapters and monographs for specific materials. Accreditation standards such as ISO/IEC 17025 address laboratory competence and traceability. Inspectors may review instrument qualification, analyst training, reference material control, and electronic records. Open questions include how best to validate methods for new complex products and how to handle automated data processing. Laboratories generally resolve these issues through risk assessment, method lifecycle management, and documented scientific justification.
In quality control laboratories, HPLC testing supports batch release, raw material checks, stability studies, and impurity profiling. A validated method defines sample preparation, instrument settings, calibration, and acceptance criteria. Analysts compare results with specifications and investigate out-of-specification outcomes before a batch is approved. Documentation includes chromatograms, integration records, audit trails, and reagent details. Because results influence product decisions, laboratories follow formal quality systems and data integrity rules. The exact tests and limits depend on the material, its intended use, and the applicable regulatory framework.
Method validation examines whether an HPLC procedure is suitable for its intended purpose. Common parameters include accuracy, precision, specificity, linearity, range, detection limit, quantification limit, and robustness. Accuracy describes closeness to a true or accepted value, while precision describes agreement among repeated measurements. Specificity shows whether the method can measure the analyte without interference from related substances. Robustness tests small deliberate changes in flow, temperature, or solvent composition. Validation is not a one-time event; methods may need partial revalidation after changes to instruments, columns, sample handling, or specification limits. Regulatory guidance provides frameworks, but some details remain method-specific.
Absinthe makes you crazy and criminal, provokes epilepsy and tuberculosis, and has killed thousands of French people. It makes a ferocious beast of the man, a martyr of the woman, and a degenerate of the infant, it disorganizes and ruins the family and menaces the future of the country.
=== Use as forage === In former times, Jerusalem artichoke was used as forage for domesticated cattle, horses, and pigs. The plant has valuable nutrient contents and various bioactive compounds, and so is used today as an animal feed source or for the health of several animal species. Pigs, for example, can eat the tuber either dried or directly from the ground or the green plant biomass (stalks and leaves) from the pasture. Washed Jerusalem artichoke tubers can be fed to many animals, and silage produced from the harvested stalks and leaves. The silage has high nutrient values and satisfactory digestion performance for ruminants. Its high inulin content beneficially affects the rumen metabolism and microflora. However, cutting the tops to produce silage greatly reduces the harvest of the tubers. There are also many other Jerusalem artichoke products on the market, such as supplementary feed for horses, dogs, and small animals.

== Applications == For the past 30 years, isothermal titration calorimetry has been used in a wide array of fields, ranging from metal binding studies to drug discovery, and nanomaterials research. In the old days, this technique was used to determine fundamental thermodynamic values for basic small molecular interactions. In recent years, ITC has been used in more industrially applicable areas, such as drug discovery and testing synthetic materials. Although it is still heavily used in fundamental chemistry, the trend has shifted over to the biological side, where label-free and buffer independent values are relatively harder to achieve.

=== Other === AMH production by the Sertoli cells of the testes remains high throughout childhood in males but declines to low levels during puberty and adult life. AMH has been shown to regulate production of sex hormones, and changing AMH levels (rising in females, falling in males) may be involved in the onset of puberty in both sexes. Functional AMH receptors have also been found to be expressed in neurons in the brains of embryonic mice, and are thought to play a role in sexually dimorphic brain development and consequent development of gender-specific behaviours. In a clade of Sebastes rockfishes in the Northwest Pacific Ocean, a duplicated copy of the AMH gene (called AMHY) is the master sex-determining gene. In vitro experiments demonstrate that the overexpression of AMHY causes female-to-male sex reversal in at least one species, S. schlegelii.
Anti-drug antibodies can cause negative side effects, accelerate the rate of drug clearance, and reduce the therapeutic effects of the biologic. In clinical practice, less than 50% of patients who showed an initial positive response to biological therapy were in remission after one year. The development of anti-drug antibodies can be reduced by limiting any times when there is no biologic present in the body and by taking other immunosuppressants (such as thiopurines or methotrexate) in combination.
. The superscript "+" indicates the ion charge and the superscript "•" indicates an unpaired electron of the radical ion. The energy of the electron beam is typically 70 electronvolts and the ionization process typically produces extensive fragmentation of the chemical bonds of the molecule. Due to the high vacuum pressure in the ionization chamber, the mean free path of molecules are varying from 10 cm to 1 km and then the fragmentations are unimolecular processes. Once the fragmentation is initiated, the electron is first excited from the site with the lowest ionization energy. Since the order of the electron energy is non-bonding electrons > pi bond electrons > sigma bond electrons, the order of ionization preference is non-bonding electrons > pi bond electrons > sigma bond electrons.
Seaport terminals handle a wide range of maritime cargoes. Break bulk / general cargo are goods that are handled and stowed piecemeal to some degree, as opposed to cargo in bulk or modern shipping containers. They are typically bundled in batches for hoisting, (with cargo nets, slings or crates), or stacked on trays, pallets or skids. They are generally lifted directly into and out of a vessel's holds, by cranes or derricks present on the dock or on the ship itself. If hoisted onto the deck instead of straight into the hold, cargo then is manhandled and stowed by stevedores. Securing break bulk and general freight inside a vessel includes the use of dunnage. When no hoisting equipment is available, break bulk would previously be man-carried on and off the ship, over a plank, or by passing via human chain. Since the 1960s, the volume of break bulk cargo has enormously declined worldwide in favour of mass adoption of containers. Bulk cargo, such as salt, oil, tallow, but also scrap metal, is usually defined as commodities that are neither on pallets nor in containers. Bulk cargoes are not handled as individual pieces, the way heavy-lift and project cargo are. Alumina, grain, gypsum, logs, and wood chips, for instance, are bulk cargoes. Bulk cargo is classified as liquid or dry.
== Safe handling == Proper handling of affected or damaged materials prevents mold from spreading. Because mold spores are released into the air, it is recommended to wear an NIOSH-rated respirator before coming into contact with affected materials. Nitrile gloves should be worn to protect the skin from contact with mold. In extreme cases, a full-body hazmat suit may be required. Materials must always be treated on a surface that can be cleaned with bleach or on neutral, disposable materials such as unprinted newsprint.

Border control policies typically require travellers to present valid travel documents to ascertain their identity, nationality, permanent residence status, and eligibility to enter a given jurisdiction. The most common form of travel document is the passport, a booklet-form identity document issued by national authorities or the governments of certain subnational territories containing an individual's personal information as well as space for the authorities of other jurisdictions to affix stamps, visas, or other permits authorising the bearer to enter, reside, or travel within their territory. Certain jurisdictions permit individuals to clear border controls using identity cards, which typically contain similar personal information.
=== Carbon dioxide === Humans are the main indoor source of carbon dioxide (CO2) in most occupied buildings. Indoor CO2 concentrations can be used as an indicator of the adequacy of outdoor air ventilation relative to occupancy and metabolic activity. Exposure to elevated indoor CO2 concentrations has been investigated in relation to human health and cognitive performance. Some studies have reported physiological and psychomotor effects at relatively low concentrations, although the evidence is not consistent and it can be difficult to distinguish the effects of CO2 itself from those of other pollutants and human bioeffluents associated with inadequate ventilation. Reviews of the available evidence have reported possible effects on cognitive performance at concentrations above 1000 ppm, particularly during complex tasks, although the evidence remains subject to uncertainty. At concentrations below 5000 ppm, the health effects attributable specifically to CO2 are difficult to distinguish from those associated with other indoor pollutants and inadequate ventilation. Indoor CO2 concentrations are widely used as an indicator of ventilation performance in buildings. Because indoor concentrations are influenced by occupancy, ventilation rates, building characteristics and outdoor conditions, measurements of CO2 can provide information on the adequacy of air exchange in occupied spaces.
The DIT and other forms of digital mass analyzers scan ions by scanning the frequency of the drive waveform. The AC voltage is typically fixed during the scan. Digital devices use a duty cycle which allows them to operate completely independent of a DC voltage and without resonant excitation. When the DC voltage is zero the parameter a is also zero. Consequently, ion stability will depend on q. With these considerations it was possible to design a new type of stability diagram that is more suitable for planning and performing experiment. In 2014 Brabeck and Reilly created a stability diagram that maps the range of stable mass-to-charge ratios, m/z to the corresponding range of drive frequencies based on several user inputs. For a particular duty cycle, the operator can quickly reference the range of stable masses at each frequency of a scan. Fig 4 (a) and (b) shows the frequency-m/z stability diagram for a linear DIT with a duty cycle of d = 0.50 and d= 0.60 respectively.
Half-Life: Alyx received "universal acclaim", according to the review aggregator website Metacritic. According to the review aggregator OpenCritic, it was recommended by 97% of critics. By April 2020, it was one of the 20 highest-rated PC games on Metacritic. VG247, TechRadar and Video Games Chronicle described it as VR's "killer app". The announcement trailer was watched more than 10 million times within the first 24 hours of its release. Though most fans expressed excitement, some were disappointed that Alyx was only available in VR, a small but growing market in 2019. Vic Hood of TechRadar expressed enthusiasm but wrote that "we forever live in hope" for Half-Life 3. Kevin Webb of Business Insider wrote that Alyx could "spark fresh interest in an industry [VR] that has struggled to win over hardcore gamers". Andrew King of USGamer suggested that Alyx would test whether the modding community would embrace VR, a difficult area to work in. In IGN, Dan Stapleton wrote that Alyx was one of the best shooter games and the best VR shooter by far. He felt that Valve "set a new bar for VR in interactivity, detail, and level design ... It feels like a game from the future, and one that the rest of VR gaming will likely take a good long while to match, much less surpass." In Polygon, Ben Kuchera wrote of how Alyx transformed FPS systems. For example, he felt that reloading guns, traditionally performed with a button press, was more fun in VR. He wrote: "The magic lies in being inside the world, being able to touch it, and interact with it, directly.
=== Quantum properties === 1H is a spin-1/2 subatomic particle and therefore a fermion. Other fermions include neutrons, electrons, and tritium. Fermions are governed by the Pauli exclusion principle, where no two particles can have the same quantum number. However, bosons like deuterium and photons, are not bound by exclusion and multiple particles can occupy the same energy state. This fundamental difference in 1H and 2H manifests in many physical properties. Integer-spin particles like deuterium follow Bose–Einstein statistics while fermions with half-integer spins follow Fermi–Dirac statistics. Wave functions that describe multiple fermions must be antisymmetric with respect to swapping particles, while boson wave functions are symmetric. Because bosons are indistinguishable and can occupy the same state, collections of bosons behave very differently than fermions at colder temperatures. As bosons are cooled and relaxed to the lowest energy state, phenomena like superfluidity and superconductivity occur.

HPLC separates and detects individual compounds in a liquid sample, producing peaks at characteristic retention times. Peak area or height can be used to estimate concentration when calibrated with known standards. It does not identify unknown compounds with certainty unless additional detectors or reference materials are used.
Pressure drives the liquid mobile phase through a column packed with small particles. Without pressure, flow would be very slow or stop because the packed bed resists liquid movement. Modern pumps maintain a steady flow despite the resistance.
A chromatogram is a plot of detector signal against time after sample injection. Each peak represents a compound or group of compounds eluting from the column. Retention time and peak area are the main measurements read from the plot.
Validation establishes suitability for a new method, while verification confirms that a method works in a specific laboratory. Verification is often used when a validated method is adopted with existing equipment and staff. Both rely on documented acceptance criteria.
