Routine quality control monitors retention time shifts, baseline noise, system pressure, and peak shape. Trends can reveal column aging, mobile phase preparation errors, detector drift, or sample degradation. Corrective actions may include replacing the column, preparing fresh mobile phase, or recalibrating the detector. Stability testing often uses HPLC to measure parent compound loss and degradation product formation. Open questions remain about how accelerated stability results extrapolate to long-term storage under varied conditions.
Quality control for HPLC testing combines scheduled checks, documented procedures, and review of results. Before sample analysis, system suitability testing confirms that the instrument, column, and method meet predefined criteria. Common criteria include resolution between critical peaks, retention time precision, peak tailing, and theoretical plate count. Failure triggers investigation before results are reported. Records link raw data, calculations, instrument logs, and analyst identity to each batch, supporting audits and repeat analysis.
System suitability testing is performed before and during analytical runs to confirm that the instrument and method are working as expected. Common checks include retention time, peak area, resolution between critical pairs, tailing factor, and theoretical plate count. Results are compared with predefined limits, and a failed check requires investigation before sample results are reported. Quality control samples at low, middle, and high concentrations are injected at intervals to monitor accuracy and precision. Blank injections detect carryover and contamination, while control charts track performance over time.
Data handling and documentation are central to HPLC quality control. Electronic systems should have audit trails that record changes to methods, sequences, and results. Integration parameters, such as peak baseline and threshold, can affect reported areas and must be defined in advance. Out-of-specification results trigger a structured investigation that may include reanalysis, instrument checks, and review of sample preparation. Regulatory inspections often examine raw data, audit trails, and training records to verify that reported results are traceable and reliable.
Method validation establishes that an HPLC procedure is suitable for its intended use. Key parameters include accuracy, precision, specificity, linearity, range, limit of detection, limit of quantitation, and robustness. Accuracy measures agreement with a true or accepted value, while precision describes repeatability and intermediate precision. Specificity confirms that the method measures the analyte without interference from impurities, degradants, or excipients. Validation is documented in a protocol and report, and acceptance criteria are set before experiments begin. Regulatory guidance varies by region, but the general principles are widely harmonized.
| Property | Value | Notes |
|---|---|---|
| Retention time RSD | ≤1% for five replicate injections | Typical criterion; method-specific limits apply. |
| Resolution | ≥1.5 between critical pair | Baseline separation is generally desired. |
| Tailing factor | ≤2.0 | Measures peak symmetry. |
| Theoretical plates | ≥2000 per column | Method-dependent; higher values indicate greater efficiency. |
| Peak area RSD | ≤2% for replicate injections | Reflects autosampler and detector precision. |
HPLC testing is an analytical technique used to separate, identify, and quantify components in a liquid sample. It relies on a pressurized mobile phase that carries the sample through a column packed with stationary phase. Different compounds travel at different rates because of interactions with the stationary and mobile phases. The resulting signal versus time is a chromatogram. Peak position indicates identity under specified conditions, while peak area or height relates to amount.
Laboratories apply HPLC testing across pharmaceutical, food, environmental, and industrial chemistry. The method can measure active ingredients, impurities, additives, preservatives, and degradation products. Sample preparation often includes dilution, filtration, and sometimes extraction or derivatization. The choice of column, mobile phase, pH, temperature, and detector depends on the analytes and matrix. Results are compared with reference standards to assign identity and concentration. Method suitability is judged by resolution, precision, and accuracy.
== Medical applications == RNA, initially deemed unsuitable for therapeutics due to its short half-life, has been made useful through advances in stabilization. Therapeutic applications arise as RNA folds into complex conformations and binds proteins, nucleic acids, and small molecules to form catalytic centers. RNA-based vaccines are thought to be easier to produce than traditional vaccines derived from killed or altered pathogens, because it can take months or years to grow and study a pathogen and determine which molecular parts to extract, inactivate, and use in a vaccine. Small molecules with conventional therapeutic properties can target RNA and DNA structures, thereby treating novel diseases. However, research is scarce on small molecules targeting RNA and approved drugs for human illness. Ribavirin, branaplam, and ataluren are currently available medications that stabilize double-stranded RNA structures and control splicing in a variety of disorders. Protein-coding mRNAs have emerged as new therapeutic candidates, with RNA replacement being particularly beneficial for brief but torrential protein expression. In vitro transcribed mRNAs (IVT-mRNA) have been used to deliver proteins for bone regeneration, pluripotency, and heart function in animal models. SiRNAs, short RNA molecules, play a crucial role in innate defense against viruses and chromatin structure. They can be artificially introduced to silence specific genes, making them valuable for gene function studies, therapeutic target validation, and drug development.
== Antithrombin and heparin == Antithrombin inactivates its physiological target enzymes, Thrombin, Factor Xa and Factor IXa with rate constants of 7–11 × 103, 2.5 × 103 M−1 s−1 and 1 × 10 M−1 s−1 respectively. The rate of antithrombin-thrombin inactivation increases to 1.5 – 4 × 107 M−1 s−1 in the presence of heparin, i.e. the reaction is accelerated 2000-4000 fold. Factor Xa inhibition is accelerated by only 500 to 1000 fold in the presence of heparin and the maximal rate constant is 10 fold lower than that of thrombin inhibition. The rate enhancement of antithrombin-Factor IXa inhibition shows an approximate 1 million fold enhancement in the presence of heparin and physiological levels of calcium. AT-III binds to a specific pentasaccharide sulfation sequence contained within the heparin polymer GlcNAc/NS(6S)-GlcA-GlcNS(3S,6S)-IdoA(2S)-GlcNS(6S) Upon binding to this pentasaccharide sequence, inhibition of protease activity is increased by heparin as a result of two distinct mechanisms. In one mechanism heparin stimulation of Factor IXa and Xa inhibition depends on a conformational change within antithrombin involving the reactive site loop and is thus allosteric. In another mechanism stimulation of thrombin inhibition depends on the formation of a ternary complex between AT-III, thrombin, and heparin.
=== Hemagglutination === Lectins are considered a major family of protein antinutrients, which are specific sugar-binding proteins exhibiting reversible carbohydrate-binding activities. Lectins are similar to antibodies in their ability to agglutinate red blood cells. Many legume seeds have been proven to contain high lectin activity, termed hemagglutination. Soybean is the most important grain legume crop in this category. Its seeds contain high activity of soybean lectins (soybean agglutinin or SBA).
=== Brust-Schiffrin method === This method was discovered by Brust and Schiffrin in the early 1990s, and can be used to produce gold nanoparticles in organic liquids that are normally not miscible with water (like toluene). It involves the reaction of a chlorauric acid solution with tetraoctylammonium bromide (TOAB) solution in toluene and sodium borohydride as an anti-coagulant and a reducing agent, respectively. Here, the gold nanoparticles will be around 5–6 nm. NaBH4 is the reducing agent, and TOAB is both the phase transfer catalyst and the stabilizing agent. TOAB does not bind to the gold nanoparticles particularly strongly, so the solution will aggregate gradually over the course of approximately two weeks. To prevent this, one can add a stronger binding agent, like a thiol (in particular, alkanethiols), which will bind to gold, producing a near-permanent solution. Alkanethiol protected gold nanoparticles can be precipitated and then redissolved. Thiols are better binding agents because there is a strong affinity for the gold-sulfur bonds that form when the two substances react with each other. Tetra-dodecanthiol is a commonly used strong binding agent to synthesize smaller particles. Some of the phase transfer agent may remain bound to the purified nanoparticles, this may affect physical properties such as solubility. In order to remove as much of this agent as possible, the nanoparticles must be further purified by soxhlet extraction.

Shortness of breath, particularly with exertion Chronic dry, hacking coughing Fatigue and weakness Chest discomfort, including chest pain Loss of appetite and rapid weight loss Pulmonary fibrosis is suggested by a history of progressive shortness of breath (dyspnea) with exertion. Sometimes fine inspiratory crackles can be heard at the lung bases on auscultation. A chest X-ray may not be abnormal, but high-resolution CT will often show abnormalities.
Although the closest counterions partially mask a charged micelle (by up to 92%), the effects of micelle charge affect the structure of the surrounding solvent at appreciable distances from the micelle. Ionic micelles influence many properties of the mixture, including its electrical conductivity. Adding salts to a colloid containing micelles can decrease the strength of electrostatic interactions and lead to the formation of larger ionic micelles. This is more accurately seen from the point of view of an effective charge in hydration of the system.

==== Fraud conviction and imprisonment ==== The PTL Club's fundraising activities between 1984 and 1987 were reported by The Charlotte Observer, eventually leading to criminal charges against Bakker. Bakker and his PTL associates sold $1,000 "lifetime memberships", entitling buyers to an annual three-night stay at a luxury hotel at Heritage USA during that period. According to the prosecution at Bakker's fraud trial, tens of thousands of memberships were sold but only one 500-room hotel was ever finished. Bakker sold "exclusive partnerships" which exceeded capacity, raising more than twice the money needed to build the hotel. Much of the money paid Heritage USA's operating expenses, and Bakker kept $3.4 million. After a 16-month federal grand jury probe, Bakker was indicted in 1988 on eight counts of mail fraud, 15 counts of wire fraud and one count of conspiracy. In 1989, after a five-week trial which began on August 28 in Charlotte, North Carolina, a jury found him guilty on all 24 counts. Judge Robert Daniel Potter sentenced Bakker to 45 years in federal prison and imposed a $500,000 fine. At the Federal Medical Center, Rochester in Rochester, Minnesota, he shared a cell with activist Lyndon LaRouche and skydiver Roger Nelson. The United States Court of Appeals for the Fourth Circuit upheld Bakker's conviction on the fraud and conspiracy charges, voided Bakker's 45-year sentence and $500,000 fine and ordered a new sentencing hearing in February 1991.
One method of feeding libraries into the microfluidic device uses single cell encapsulation, in which droplets contain a maximum of one cell each. This avoids confounding results that could be generated by having multiple cells, and consequently multiple genotypes, in a single droplet, while maximizing the efficiency of resource consumption. This method enables the detection of secreted proteins and proteins on the cell membrane. The addition of a cell lysate to the droplets, which breaks down the cellular membrane such that the intracellular species are freely available within the droplet, expands the capabilities of the single cell encapsulation method to analyze intracellular proteins. The library can also be made entirely in vitro (i.e., not in its biological/cellular context) such that the content of the droplet is exclusively a mutated DNA strand. The in vitro system requires PCR and the use of in vitro transcription and translation (IVTT) systems to generate the desired protein in the droplet for analysis. Sorting of droplets for directed evolution is primarily done by fluorescence detection (e.g., fluorescence-activated droplet sorting (FADS)), however recent developments in a absorbance-based sorting methods, known as absorbance-activated droplet sorting (AADS), have expanded the diversity of substrates that can undergo directed evolution through a droplet-based microfluidic device. Recently, sorting capability has even expanded to the detection of NADPH levels and has been used to create higher activity NADP-dependent oxidoreductases.

The Hukbong Bayan Laban sa Hapon (lit. 'People's Army Against the Japanese'), better known by the abbreviation Hukbalahap, was a Filipino communist guerrilla movement formed by the farmers of Central Luzon. They were originally formed to fight the Japanese, but extended their fight into a rebellion against the Philippine government, known as the Hukbalahap rebellion in 1946. It was eventually put down through a series of reforms and military victories by Defense Secretary, and later President, Ramon Magsaysay. Constituted in March 1942, the Hukbalahap was to be part of a broad united front resistance to the Japanese occupation of the Philippines. This original intent is reflected in its name. By 1950, the Communist Party of the Philippines (PKP) had resolved to reconstitute the organization as the armed wing of a revolutionary party, prompting a change in the official name to Hukbong Mapagpalaya ng Bayan, likely in emulation of the Chinese People's Liberation Army. Notwithstanding this name change, the HMB continued to be popularly known as the Hukbalahap, and the English-speaking press continued to refer to it and its members, interchangeably, as the Huks (Singular: Huk) during the whole period between 1945 and 1952. A monument dedicated to the Huks in Cabiao, Nueva Ecija, was constructed to honor their actions during World War II. On October 14, 2023, the Luis M.

System suitability is typically performed before each batch or according to the validated method and laboratory procedure. Some long runs include periodic checks during analysis. The required frequency depends on regulatory expectations and method performance.
Retention time drift can result from changes in mobile phase composition, column temperature, pump flow, or column age. A gradual shift often points to column degradation. A sudden shift may indicate a leak, mixing error, or incorrect mobile phase.
Retention time alone cannot confirm identity because different compounds may elute at similar times. Coupling HPLC with mass spectrometry or comparing against authenticated standards increases confidence. Confirmation usually requires orthogonal data.
System suitability is a set of checks that confirm the instrument and method perform within limits before sample analysis. It typically includes resolution, tailing factor, retention time, and peak area reproducibility. If a check fails, the run is invalidated until the cause is resolved.
